CL-316243: Repeat Run

CL-316243 previously produced a clear visual reduction in body fat. Most of the change happened in the first two weeks, then slowed. This run asks whether that useful early response repeats. Personal experiment record, not medical advice, prescribing, sourcing guidance, or an instruction to copy.

Primary question: Does the earlier visual fat-loss result repeat, and does the response slow again after the first two weeks?

Added to the active ledger on September 10, 2026. The recorded schedule is 5 mg every morning while fasted, also listed in the Daily Stack. The focus is the change in body composition across the run.

The first run was helpful

The completed August record covers approximately one month of use. I saw less body fat in the mirror, especially during the first two weeks. I did not feel an acute effect around training. The run closed as helpful, and I removed CL while moving on to SANA. That result remains intact.

How the repeat will be read

Standardized visual comparisons are the main readout, supported by waist measurements and the seven-day body-weight trend. The early comparison is the first two weeks against the starting record; later comparisons will show whether the response continues, plateaus, or changes direction. Similar lighting, posture, time of day, and measurement conditions make the visual record easier to read.

Session energy and training output remain useful context, but a stimulant-like feeling is not required for a helpful result. Resting heart rate, blood pressure, temperature, sleep, and unusual fatigue will sit alongside the body-composition observations.

The routine around this run

This repeat takes place alongside ATX-304, SANA, BAM15, Retatrutide, and the ongoing cut and reverse diet. The record will keep their changes, calorie intake, and training dates visible. The practical question is whether the earlier response returns in this routine. The new run is active; a repeat result has not yet been recorded.

Research Summary

Beta-3 signaling and fat metabolism

CL-316243 is a selective beta-3 adrenergic receptor agonist. In high-fat-fed rats, chronic treatment increased resting energy expenditure and body temperature, reduced body fat, and increased brown-fat uncoupling-protein content. That is the foundation of the thermogenic interest, with the animal findings kept separate from the direct human data. Himms-Hagen et al., 1994.

What the human trial actually found

In an eight-week trial, 14 lean young men were randomized to CL or placebo. At week eight, the CL group had 23% greater fat oxidation, but no increase in 24-hour energy expenditure and no change in body weight or composition. The study therefore demonstrates a shift in fuel use. My earlier visual fat-loss result is recorded separately. Weyer et al., 1998.

Why the early response is worth recording

Insulin-mediated glucose disposal improved at week four in that trial, then weakened as measured CL concentrations declined by week eight. This gives a useful research question about response over time, but it does not identify the cause of my first run's slowdown. The repeat will record when the visible change occurs without automatically labeling a plateau as receptor tolerance.

Exposure and tolerability context

The human study's 1,500 mg/day regimen is not an exposure match for this record. It does not validate 5 mg or morning-fasted timing. No heart-rate or blood-pressure effect or tremor was reported in its small sample; that is a limited observation, not a long-term safety determination.
